Physical and Chemical Considerations in Microwave Assisted Peptide Synthesis: Method Development to Automated cGMP Peptide Production
Abstract
To address the needs of the market, new cGMP methodology has been developed utilizing automation and microwave assisted heating. This work details mechanistic-based, innovative improvements to the chemical methodology of solid phase peptide synthesis and addresses high temperature considerations such as aspartimide formation, epimerization, and heat-sensitive linkers (ex 2Cl trt PS). Key features include: (1) enhanced methods for high purity peptide synthesis, (2) green methodology with a demonstrated 95% reduction in solvent consumption, and (3) rapid scale up from R&D to production with cGMP considerations.
